Design and Simulation of a Microscale Magnetophoretic Device for the Separation of Nucleated Fetal Red Blood Cells from Maternal Blood
نویسندگان
چکیده
Intense research has been carried out into methods that aim at harvesting fetal cells from maternal blood as substitutes to amniocentesis and chorionic villus sampling. Although much has been accomplished using well established techniques, a rapid and inexpensive method to separate fetal cells with great accuracy and efficiency to maximize cell yield is still required. This work focuses on the separation of fetal nucleated red blood cells from the maternal circulation based on their intrinsic magnetic properties. The design and simulation of a magnetophoretic separator is described, as it will be one of the stages of a lab-on-chip for non-invasive prenatal diagnosis (NIPD). Finite Element Analyses have been used to assist design choices and to confirm analytical results. A final layout has been validated for manufacturing and testing. Successful fabrication and positive test results of the device will constitute a further step towards the implementation of new techniques for NIPD.

Background: The presence of increased numbers of nucleated red blood cell (NRBC) in the umbilical cord blood has been associated with states of relative hypoxia. Nucleated red blood cell counts are a potentially useful tool in estimating the degree and timing of intrauterine hypoxia. This may have important implication in determining causality in case of compromised infant. Cord blood NRBC coun...
متن کاملI-40: Non Invasive Prenatal Genetic Diagnosis;Current Status and The Future
Discovery of cell free fetal DNA in 1997 has deeply changed the outlook of prenatal diagnosis approaches as most of the clinically established screening tests are not sensitive/specific enough while the current practical diagnostic tests are also invasive in their nature. The most common prenatal screening test is routinely practiced for the diagnosis of Down syndrome (DS) which includes a 10% ...
متن کاملInvestigation of Changes in Nucleated Red Blood Cells in Neonatal Infection
Background: Early diagnosis is paramount to the treatment of neonatal infections. Nucleated red blood cells (NRBCs) are immature erythrocytes that increase in number due to stress and hypoxia. This study was conducted to determine NRBC count in the peripheral blood and examine its significance as a marker of neonatal infection. Methods: This case-control study was conducted on 154 infants (78 ...

This paper presents the characterization of a continuous magnetophoretic microseparator for separating white and red blood cells from whole blood by using a high gradient magnetic separation method and microfabrication technology. The magnetophoretic microseparator directly separates blood cells based on their native magnetic properties without the use of additives such as magnetic tagging or i...
